Les biochimistes et les biologistes consacrent souvent des jours, voire des semaines, \u00e0 la pr\u00e9paration et \u00e0 la purification de quelques centaines de microlitres d&#039;\u00e9chantillons pr\u00e9cieux.<\/p>\n<p>Il est donc logique que leur temps soit mieux employ\u00e9 si ce processus est rendu aussi efficace que possible.<\/p>\n<p>Auparavant, l&#039;id\u00e9e de r\u00e9aliser une s\u00e9rie compl\u00e8te d&#039;exp\u00e9riences avec seulement 100 \u00b5l de solution \u00e9tait consid\u00e9r\u00e9e comme une utopie, car de tels volumes \u00e9taient n\u00e9cessaires uniquement pour amorcer l&#039;instrument.<\/p>\n<h3><strong>100 \u00b5l de solution m\u00e8re, 10 tirs, temps mort de 1 ms<\/strong><\/h3>\n<p>La r\u00e9duction de la consommation d&#039;\u00e9chantillon dans les instruments \u00e0 flux arr\u00eat\u00e9 a \u00e9t\u00e9 notre principale motivation pour le d\u00e9veloppement du \u00b5SFM. Pas une seule goutte de liquide pr\u00e9cieux n&#039;est gaspill\u00e9e, ni dans le flux arr\u00eat\u00e9, ni dans le flacon de stockage. Avec seulement 100 \u00b5l de solution m\u00e8re, l&#039;utilisateur dispose de suffisamment de solution pour r\u00e9aliser 10 r\u00e9plicats. Ce nombre peut m\u00eame \u00eatre \u00e9tendu \u00e0 25 r\u00e9plicats lors d&#039;exp\u00e9riences de repliement\/repliement\u00a0! Ces performances sont obtenues avec notre microcuvette standard, sans perte de sensibilit\u00e9 et avec un temps mort de seulement 1 ms.<\/p>\n<h3><strong>Allier pr\u00e9cision et modularit\u00e9<\/strong><\/h3>\n<p>Le \u00b5SFM est un syst\u00e8me \u00e0 flux arr\u00eat\u00e9 \u00e0 deux seringues. Chaque seringue est actionn\u00e9e par un moteur pas \u00e0 pas ind\u00e9pendant, ce qui permet de modifier librement le rapport de m\u00e9lange de 1:1 \u00e0 1:9 via le logiciel Bio-Kine. L&#039;association de moteurs de haute pr\u00e9cision et de seringues en verre de microvolume permet d&#039;injecter quelques \u00b5l d&#039;\u00e9chantillons pr\u00e9cieux \u00e0 chaque exp\u00e9rience.
